《科学》子刊:“我死两次”!科学家首次发现中性粒细胞胞外诱捕网真相,为灭外敌它竟连续“自杀”两次,太悲壮了丨科学大发现2023-12-30 19:14·奇点网*仅供医学专业人士阅读参考中性粒细胞是最积极、最强力的免疫细胞之一。在发生感染之后,它是第一批从血液中赶到发病组织的免疫细胞。在对抗病原物的过程中,中性粒细胞也非常凶狠,可以吞噬病原物,可以释放活性氧毒杀病原物,甚至还会通过自身的死亡来阻止病原物。中性粒细胞会通过“自杀”对抗病原物这一现象,是科学家在2004年首次发现的[1]。简单来说,在面对病原微生物的时候,活化的中性粒细胞会将核DNA释放到细胞外,通过中性粒细胞胞外诱捕网(NET)捕获并中和病原物[2]。中性粒细胞胞外诱捕网示意图[2]既然DNA都释放出去了,中性粒细胞肯定就死了,科学家把中性粒细胞的这种程序性死亡方式叫做NETosis[2]。一直以来,科学家都认为,NETosis和细胞凋亡是两个独立的死亡过程。然而,由加州大学圣地亚哥分校Ben A. Croker和奥古斯塔大学Yangfang "Peipei" Zhu等领衔的研究团队,近日在著名期刊《科学·进展》上发表了一篇重要研究论文[3],挑战了我们对细胞死亡的认知。他们的研究表明,中性粒细胞的凋亡和NETosis不是毫无关系,而是两个连续发生的死亡过程。简单来说,中性粒细胞的凋亡会进一步引发NETosis。据了解,这也是科学家首次发现凋亡与NETosis之间存在关联。值得注意的是,在结合之前的研究成果之后,研究人员发现,在中性粒细胞中,多种死亡途径(焦亡、坏死)都汇聚于NETosis。由于中性粒细胞的NET与肿瘤和一些自身免疫疾病有关,这一发现可能带来新的治疗思路。论文首页截图细胞凋亡,大家一定都不陌生了。它是一种重要的程序性死亡方式,对于维持机体的稳定非常重要。NETosis从发现至今也有近20年的历史了,科学家已经知道了它的发生过程。简单来说,在中性粒细胞中,打孔蛋白GSDMD(参与细胞焦亡)和MLKL(参与细胞坏死)会在中性粒细胞膜上打孔,导致钙离子流入细胞内;随后,肽酰基精氨酸脱亚氨酶4(PAD4)就会被激活,而激活的PAD4会将组蛋白瓜氨酸化;这种变化会导致DNA从组蛋白上解旋,随后DNA被挤到细胞外,形成NET,捕获病原物[4]。中性粒细胞的两次死亡为了探索细胞凋亡与NETosis之间的关系,Croker团队首先用多种诱导凋亡物质刺激中性粒细胞。结果他们发现组蛋白普遍被瓜氨酸化,这表明细胞凋亡与NETosis之间可能存在联系。在敲除中性粒细胞的PAD4之后,组蛋白的瓜氨酸化现象就消失了。以上研究说明,凋亡确实与NETosis密切相关。他们还通过染色的方法,从中性粒细胞的形态学变化上证实了上述发现。至于背后的分子机制,Croker团队发现,凋亡的蛋白酶Caspases可裂解并激活打孔分子GSDME,这导致钙离子流入细胞内,诱发了NETosis。如果敲除编码GSDME的基因,中性粒细胞虽然还可以启动细胞凋亡,但发生NETosis的能力大幅减弱。Ben A.
